Well, that clarified things.

I moved my .sylpheed-2.0 and Mail directories to another directory and
ran Sylpheed (3.5.0beta1). It gave me a choice where to make the mh
directory; I took the default (~/Mail).

Looking at the new .sylpheed-2.0 directory, I found in the file
folderlist.xml the following:

So you can either start Sylpheed's set-up process as I did above and
then rebuild the folder tree, or edit ~/.sylpheed-2.0/folderlist.xml to
change the path to what is shown in the second line of the file as listed
above and then rebuild the folder tree.
