The Sylpheed readme.txt file says:

"If you want to register Sylpheed as a handler for mailto: protocol after you
have installed it from zip archive, add the entry to the registry by
double-clicking the included file "sylpheed-mailto-protocol.reg" (you can
choose it while installing by the installer). If Sylpheed is installed into
a location other than C:\Program Files\Sylpheed\sylpheed.exe, modify the path."
