  Nope, that's not possible currently.

  Adding a specific existing functionality is far easier, the attached patch
  demonstrates that for mark as read function, you can add others. It's
  using an apply stock icon and is not enabled by default (you have to
  customize mail toolbar first), so I guess there's room for improvement.
  As a side effect it seems to work as expected ;)
